Disorders of GNAS inactivation comprise a clinically heterogeneous spectrum characterized by varying degrees of hormone resistance, features of Albright hereditary osteodystrophy, and ectopic ossification. They result from genetic or epigenetic defects affecting the imprinted GNAS locus, with disease manifestations strongly influenced by the parental origin of the affected allele.
