Overview
Monogenic obesity is a rare, Mendelian form of severe early-onset obesity. It is associated with germline pathogenic variants in single genes of large effect, most affecting hypothalamic pathways involved in appetite regulation. While distinct from common polygenic obesity, which results from the cumulative effects of numerous genetic and environmental factors, it comprises a genetically and clinically heterogeneous group of disorders that includes both non-syndromic and syndromic forms.
