PGx 5-fluorouracil
Fluorouracil (5-FU) and its oral prodrug capecitabine are fluoropyrimidine cytostatics used across many solid tumours. Dihydropyrimidine dehydrogenase (DPD), encoded by DPYD, is the rate-limiting enzyme in their catabolism, and reduced or absent DPD activity predisposes to severe, potentially fatal treatment toxicity. This analysis genotypes the four consensus DPYD variants used to assess DPD deficiency before fluoropyrimidine therapy and reports the derived metaboliser status.
