PGx siponimod
Siponimod is used to treat active secondary progressive multiple sclerosis, with relapses or imaging features of inflammatory activity. It is metabolised principally by the polymorphic enzyme CYP2C9, so its systemic exposure depends strongly on CYP2C9 genotype. The EU product information therefore requires CYP2C9 genotyping before treatment is started, because siponimod is contraindicated for one CYP2C9 genotype and its use in others is subject to a genotype-dependent restriction.
