DROPLET DIGITAL PCR

Droplet digital PCR (ddPCR) is a cutting-edge technique in genetic diagnostics, offering precise quantification of DNA and RNA. Renowned for its exceptional sensitivity and accuracy, ddPCR works by distributing DNA or RNA molecules into thousands of tiny reaction chambers before amplification, with the aim of obtaining only a single target molecule per reaction chamber. This is followed by an endpoint PCR process using fluorescent labeled probes. The resulting fluorescence signals allow for highly accurate quantification. The compartmentalization into numerous reaction chambers enables absolute quantification by measuring each reaction chamber independently and correlating the number of positive reactions directly with the initial quantity of target molecules in the sample.

ddPCR is invaluable in genetic diagnostics, as it detects genetic alterations at the molecular level. This capability is critical for identifying clinically relevant changes and supporting personalized medical treatments.

Applications of ddPCR:

In cancer research, it enables the detection of rare variants, fusions and alterations in tumor tissues, which is crucial for early diagnosis and tailoring personalized therapy.

In infectious diseases, it is used to quantitatively determine the viral load in the blood and to monitor the effectiveness of therapy.

In genetics and heredity studies, it enables the detection and quantification of specific genetic variants in DNA samples, aiding in understanding inherited conditions.

ddPCR offers significant advantages, including accurate quantitative results, high sensitivity for the detection of rare genetic variants, and minimal interference from PCR inhibitors or background DNA. Potential challenges or limitations include cost, complexity, and throughput, as ddPCR can be more laborious than conventional PCR and requires specialized equipment and expertise. Nevertheless, the advantages of ddPCR and its potential for advancing genetic diagnostics and personalized medicine outweigh these challenges.
